Principle
The principle of spectrophotometry analysis is to use the phenomenon of selective absorption of light of different wavelengths by substances to conduct qualitative and quantitative analysis of substances. Through the analysis of absorption spectra, the structure and chemical composition of substances can be determined. Using a prism or grating to obtain monochromatic light, the monochromatic light continuously passes through the solution in sequence, and the absorption of the solution at each wavelength is measured to obtain the absorption spectrum curve.
This instrument operates based on the principle of relative measurement, which involves selecting a solvent (distilled water, air, or sample) as a reference solution and setting its transmittance (i.e. transmittance T) to 100%. The transmittance of the tested sample is obtained relative to the reference solution. The change in transmittance (transmittance T) has a certain functional relationship with the concentration of the measured substance, and within a certain range, it conforms to the Lambert Beer law.
